The pretty little instrumental on Eat a Peach -- is that one guitar or two?

It's two, although you can play it with one. Dickey Betts is just playing a few root notes as best I can tell.
Incidentally, Berry Oakley also played on the song, but his bass was removed from the mix when Eat a Peach was released. When the Allmans end a concert these days, they play "Little Martha" over the PA system with Berry's part restored, which makes it a nice tribute to him and to Duane.

A tab website I found said that Duane is playing in open E and Dickie is playing in standard tuning. I think playing both parts on one guitar should be easy if you're Leo Kottke.

Yeah -- I'd always thought it was two, but then when I checked AMG and saw that Leo Kottke covered it, I wasn't sure. After all, it's not like he's any kind of amazing 12-string guy or anything... ;)
