In _Dr. Horrible’s Sing-Along Blog_, the Bad Horse Cowboys are played by the three Whedon brothers.

And I once watched a director (unknowingly) call up a brother-and-sister pair to read for Romeo and Juliet at an audition. Wonderful results.
