The Life of Riley was a popular early television show starring William Bendix as Riley. But the first season had Jackie Gleason playing the character. And before that, in 1948, two live pilot programs were aired, one starring Herb Vigran and one starring Buddy Gray.
But before that, the show ran on radio for many years, with Riley’s voice being… William Bendix, who also starred in the 1949 movie.

Caity Lotz has played Sara Lance on Arrow and then Legends of Tomorrow for almost ten years. I’d totally forgotten, but in the character’s first appearance, she was played by Jacqueline MacInnes Wood, who declined to continue when the character was made a regular.

What I meant was that Norton originated the role within the MCU, much like Howard did for War Machine. Otherwise, it could also be pointed out that James Avery portrayed Rhodie in the '90s Iron Man cartoon long before Howard and Cheadle did.
Sharon Gless replaced the original Cagney on Cagney & Lacey, after Meg Foster in the pilot episode was deemed too butch for a show about 2 policewomen partners.
And Meg Foster was cast after Loretta Swit played Cagney in the original TV movie.
Phyllis Coates played Lois Lane in The Adventures of Superman during its first season, before being replaced by the better-remembered Noel Neill. But Neill actually played Lane in the 1948 Superman movie serials. Of course, neither of them originated the role - that was actress Joan Alexander on the radio series.

Most people think Peter Graves is the head of the IMF in Mission: Impossible, but Stephen Hill originated the role. Different names, but Hill’s Dan Briggs is often forgotten.

In my mind Norton is the only actor I associate with MCU Banner, since he so perfectly captured the character. I actually had to scroll up and reread your post to remember Ruffalo’s name.
I think many people still associate The Doctor with Tom Baker or David Tennant.

Audrey Meadows is the Alice Kramden we all remember from The Honeymooners. She’s also the only cast member to receive residuals from the show (thanks to her shrewd manager).
But the original Alice was played by Pert Kelton back when The Honeymooners was still a sketch routine on The Jackie Gleason Show (1952). She was replaced after being blacklisted. At first Gleason didn’t want Meadows cast because he felt she was too young and pretty for the part of his wife. So, Meadows uglified herself for her next audition and nailed the part.
