I would think that Jell-o would dry out pretty quickly…
It would be worth a try - and if it works, a more permanent version could be made with two-part silicone.
How about specially designed glasses? The stem goes up through the glass and the top where it attached is sealed. When you hang the glass from the stem, the opening is now at the top.
