Depends on ‘what for’ really.
Visually, Inara reminds me of a girl I dated for a while. Plus she has a lot of personality traits I just find appealing. I’d have no objection to her as a life partner in some quiet corner of the galaxy.
River is probably the smartest, deadliest, spookiest phenomenon in the 'verse and Summer Glau is just super-sleek and sexy (IMHO). I think together we could conquer the 'verse – or at least rattle the Alliance’s cage for a decade or so.

I voted but it needs to be said for the record that all of them are a good choice.
I find it interesting though that the favorite woman on shows like this are rarely the ostensibly super hot chick (in this case Inara). So many guys prefer Velma to Daphne or Mary Ann to Ginger and, in this case, Kaylee to Inara.
Since we’re allowed to use whatever criteria we like, here are some more:
Conversation about art, music and culture: Inara, of course.
Organizing the office awayday: Zoe, if only because Kaylee would want to do all those “fun” teambuilding exercises that I can’t stand.
Doing stand-up at open mike comedy night: River. It’d be so surreal it would go from funny to awkward and right out the other side to funny again.
Mixed doubles partner: Zoe. She’s probably got a serve like a bullet train. Kaylee and Inara don’t seem that sporty and River would probably get distracted and start a conversation with her tennis racket or kill the opposing players with the ball.
Watching old Looney Toons cartoons on the couch in sweats while eating breakfast cereal even though it’s two o’clock on a Sunday afternoon: Kaylee. She seems like she’d be a Bugs Bunny fan.
Poker night: Zoe, Inara and Kaylee. River would constantly disrupt the game by saying and doing weird shit and then would read everyone’s minds and win.
Hire as a catsitter: Kaylee.
Elect President: Either Zoe or Inara, depending on whether I felt we needed someone with a better grasp of hard power or soft power at the time.

Canoeing vacation in the Adirondacks: I don’t know why but I want to say River. I’d likely regret it at the time (and get dumped into the water more than once) but afterwards it will all be fondly remembered.
Yahtzee games night: Inara. She’s the only one that could making playing Yahtzee bearable.
Shopping trip to Bed, Bath and Beyond: Zoe, for a no-nonsense approach. Go with Kaylee and you will be hours looking at stupid cute little gewgaws and will come home with a bag of scented candles.
Building a shed: Kaylee, on the assumption that her technical skills are transferable.
Stuck in an elevator with: Inara. The best conversationalist of the lot.
Air hockey: Kaylee. River would score a shut out in less than 15 seconds, Zoe would be too competitive for it to be fun, and Inara would let me win. Kaylee is probably on my skill level and would enjoy it, win or lose.
Pub crawl: All of them. It’d be a big laugh as long as nobody plays the Fruity Oaty Bars theme. And you *know *you want to see how much River can drink and still play beer pong.
I do not want to be anywhere in the same zip code as a drunk River.
Or planet, for that matter.
I know why; a canoeing vacation without a river is really more trouble than it’s worth.
