I’ll jump in to this thread and mention Psychostick. Their song “Beer!” was the #1 song on satelite radio’s Liquid Metal for a few weeks. “Two Ton Paperweight” is also hilarious.

Mashup band Rock Sugar - the “backstory” is that a hair band from the ‘80s was marooned on an island with only a young teen girl’s music collection to listen to. Result? Mashup songs like Don’t Stop the Sandman - Journey’s "Don’t Stop Believin’" plus Metallica’s “Enter Sandman.” Jess Harnell is the lead singer; he’s probably better known to most people for the voice work he does, especially the Animaniacs’ Wakko Warner.
You may like Aussie band TISM whose details are far too involved to attempt to summarize. Here is (He’ll Never Be An) Old Man River more commonly known as I’m On The Drug That Killed River Phoenix as the title never appears in the song.

I should probably be deeply offended by “Eskimo”. The only problem with that is that the first time I heard it I almost had to pull over so I didn’t hit anyone, I was laughing so damn hard.
The three survivng members got back together in 2008 with Dan Stevens replaced the late Dave Blood. They just had a show in Boston on June 12.
Have to second this - if you like Lonely Island and Tenacious D you absolutely need to check out Jon Lajoie.

Three Dead Trolls in a Baggie are truly awesome, although a lot of their stuff may be too Canadian for a wider audience. Their website is considerably less awesome and I don’t find much of their awesomeness there, not clear if it’s because they’ve neglected to post it or if it’s just because their website is so awful. Here’s a video of a performance of Mom’s Coming to My House for the benefit and illumination of high school students, because (seriously) we just don’t give a fuck up here. (Not Giving a Fuck - try some today!)

I liked it initially, but I don’t see him having much replay value for me.
So far, my two favorites to come out of this thread have been Garfunkel & Oates (I already loved Kate Miccuci), and Rock Sugar.
