Been a while since I read it, but Demosthenes was clearly his sister.
According to the World History web site,
It’s been a while since I read Ender’s Game and its sequels, but IIRC, Ender’s sister (Valerie?) was really PO’d that she had to be Demosthenes and her brother (Peter?) got to be Locke. I guess Demosthenes (the original one, I mean) wasn’t too popular after King Philip prevailed.

Valentine, Ender’s sister was unhappy because (noting that I am not familiar with the works of either Demosthenes or Locke) she felt that her personality was better suited to Locke, while Peter’s was better suited to Demosthenes. Peter originally made them switch because he didn’t think Valentine could write as Demosthenes without his help, and he didn’t want her to become independent. She was a pacifist, and he was overly aggressive.
And BTW, there’s another Ender book out: Ender’s Shadow. It’s set at the same time as Ender’s Game, but it’s from the point of view of Bean.
