Oddly enough, quote tags do hide the text from the mouse-over preview, so any spoilers in your OP will be safe if you do this.

FWIW, I think this is because the quote tag is a built-in feature of vBulletin, so it just ignores quotes because they usually don’t help determine the content of a thread. But I believe the spoiler tag is an add-on, so the board software doesn’t know to keep it out of the previews.
Of course, I could be wrong.
