It is an internally consistent even if specious argument.
The charge is that she “plays the Black card” when it is useful for her, including getting hired for sake of the identity rather than for qualifications. Meanwhile culturally she was raised by her South Asian medical researcher mother, spent years 12 to 18 in Montreal, and married a white Jewish man. Her embrace of American Black culture is most evidenced by the majority of her college experience being at Howard, her lasting association with her Black sorority, and a strong Black mother figure in her neighborhood as a child.
As long as the trumpeting of her as a Black woman is coming from others and by way of her association with and acceptance those Black organizations the charge will I think land flat or mostly backfire. Better to let others call her Black even in an attempt to rev up the racist votes, than for her to raise it much herself. I think.
Personally, I think the best reply would be “People can have more than one aspect to their identity. For example, I am Indian, Jamaican, and African-American, while my opponent is a convicted felon, a rapist, and a moron”.
I’m not sure if she would gain anything by dignifying his argument although it could be worthwhile to wonder out loud why exactly it is that Donald Trump thinks racial purity tests are so important.
So, let’s say Kamala has 1/4 African heritage (quite frankly, I don’t know, and I don’t care). Before the Emancipation Proclamation, that was more than high enough percentage for her to be enslaved. Wasn’t it a “rule” in the Jim Crow south that a drop of African blood makes you black? If these people are so invested in bringing us back to those times, then why are they complaining about her being called “black”?
Donald, you didn’t “know” she was black because you didn’t care. She didn’t hide it; she is proud of attending Howard and belonging to her sorority. She has been active in both South Asian and African American causes. Just because you didn’t notice doesn’t remove the fact.
The way trump keeps (almost certainly on purpose) mispronouncing ‘Kamala’ is another way she can slam him.

Just to make it easier for you, just call me Ms. Harris or Madame Vice President from now on. It will be less embarrassing for you than constantly getting my name wrong. You don’t want people to think you’re stupid, right?”

“So I don’t know - Is she Vice President? Or is she President?”
If Trump’s people decide that they, too, would like to invent attacks based on Buffy the Vampire Slayer eps, then I’d recommend they accuse Kamala of being an Inca Mummy Girl. You know, to further mix up her ethnic-heritage image.
(Of course it will backfire by bringing the Democrats the South-American-heritage vote, which is a fairly large bloc.)

Harris is not in charge of border security or immigration policy. She was tasked with addressing the root causes of migration from three Central American countries.
In defending his “childless cat lady” comments, Republican vice presidential nominee JD Vance wrongly claimed that Vice President Kamala Harris “is calling for an end to the child tax credit.” He also incorrectly claimed that Harris said “it was a bad idea to have kids because of climate change anxiety.”
