He hasn’t actually said yet that it was a TV show, so don’t assume that it is.
Whatever it is, it appears that it’s an event that stopped occurring when the students were at most infants (hence why freshmen wouldn’t make the jokes), but that it’s nonetheless one that all of the students were familiar with (else they wouldn’t make the joke at all). Is this correct?

Presumably, it actually means when the parents had sex. The gametes merging can be up to a few days after that.

Was this some event (or lack of event) that happened for 3 years only, on Thursday nights, to coincide with the birth years of the sophomores, juniors, and seniors?
Would this joke work in other nearby towns? Other states in the US? Other countries?

I’m guessing the answer is No then. “Must See TV Thursday” was started in the 80s by NBC? I think. So there would be some dividing line of conception before and after Must See TV. If you are not watching TV, you might be getting frisk-eee. It was a stretch.

Dude, I was just being facetious. There’s lots of euphemisms for sex, like “did they form the beast with two backs,” “did they make whoopee,” “did they rut like rats” and so forth. I wasn’t teaching biology class. I thought that would be obvious.
