As to #2, I found this in the movie description on Wiki:
The title of the film is a reference to the 1941 Preston Sturges film Sullivan’s Travels, in which the protagonist is a director who wants to film O Brother, Where Art Thou?, a fictitious book about the Great Depression.
No mention of a line in Homer’s Odyssey, although the plot is loosely based on that.

It probably depends on the translation you read. When I read it, it was a modern translation without words like “art,” and because it has lots of figurative language that refers to family. It wouldn’t surprise me if there is a line that means “this is unfamiliar to me,” but uses some kind of metaphor of “brother,” and is translated “Brother, where art thou?” by one author, and “This is not known to me” by another. I know there are passages in the Torah in Hebrew that come out all kinds of different ways in English,
Speaking of the Torah, the title is also an inversion of Genesis 4:9, where God asks Cain “where is your brother?”
