I hope these haven’t already been mentioned:
Piled Higher and Deeper
I like Monty.
Keep reading, at least until the Jagermonsters show up. It’s a hilarious comic. Also, it wasn’t originally a webcomic: you didn’t read the first five or six strips, you read the first five or six pages of the first issue. The first part of the story was written with the expectation that you’d have a whole comic book in front of you, and was paced with that in mind. Foglio didn’t try to bring the funny on every page, because he had the whole book to work in each element of what his story was going to be about.
I like Tom the Dancing Bug.
Why don’t you run down to your local comic-book store and pick up the keenspot promotional free comic book day album? Lots of samples from webcomics there.
What, no love for Goats? Granted that the current strips are part of a vast storyline involving interdimentional pubs and demons and the Mayan calendar and Woody Allen’s laptop, but the earlier strips are gag-a-day and short storylines.
Plus, Rosenberg makes the best t-shirts.
Achewood! The dialogue is so damn catchy you’ll find yourself quoting it without realising. Make sure you read through the archives, though, as there’s some fairly dense character development.
Another vote for Perry Bible Fellowship. Here are my some of my favorites:
I’m also going to recommend The Parking Lot is Full before somebody else does. They stopped making 'em a while ago but the scroll through the archives and you’ll find some gems.
Neither of them have continuing story lines though. =|
Ex-doper Jin Wicked does a great comic: Crap I Drew on my Lunch Break
YAAAY!
Her strip is back!!!

It’s fairly casual humour, not really mind-intensive like most (What’s that comic which has the tagline “humorous comic about personae” and really fancy wording for previous/next? Yeah, that one) but still all around fun. Singularly game-oriented, with just a few exceptions.
Day by Day. Political.
Errant Story. Very stylish fantasy, in the anime mould.
Kevin & Kell. A mixed family of Furries.
Help desk. A riff on Microsoft.
Alas Pibgorn is no more.
For those readers who want a link to the Order of the Stick, here you go.
This is my personal favorite, it’s done 5 nights a week and pretty damn funny - Questionable Content (comic is SFW)

Don’t forget to read the mouse-over text for each strip.
People already beat me to some of the ones that occurred to me (Erfworld, Goblins , the Casey & Andy archives); I’ll add Nodwick and Full Frontal Nerdity.
Also Dork Tower (I’m pretty sure this is the right link, but the site seems to be down at the moment).
It shall return. Brooke McEldowney, the creator, is looking for a new place to host it where he has full creative control. He has a blog to keep in touch with us.
In addition to the previously mentioned Order of the Stick (which I love!), I also enjoy Two Lumps , the adventures of two Russian Blue cats.
I am required by the terms of my unpaid job there to also recommend Sluggy. It isn’t laugh out loud every day funny, it’s more of a graphic novel in strip form. It can be dramatic but it focuses on humor. It’s best to start reading from the beginning (almost 10 years of archives!) or start at the beginning to get some basic character info then jump to the most recent storyline. Stop by the Sluggy Related Chat forum on the message boards if you have any questions about what’s what.
I also like Something Positive but got out of the habit when I lost my cable connection.
