I think Ghostbusters, which has already been mentioned by Biggirl, is my favorite New York movie, though.

Dog Day Afternoon and Ghostbusters (“Why worry, each of us is wearing an unlicensed nuclear accelerator on his back.”) are two of my favorites also. But, this thread would be incomplete without Goodfellas, A Bronx Tale, and the enduring The Godfather parts I & II
Yeah, I’d nominate Ghostbusters as well. Any city whose citizens show up and cheer at the potential end of the world gets my vote.
I always thought “Working Girl” caught a little of the zietgeist of 1980s New York. Carly Simon’s video of “Let the River Flow” has some lovely New York scenes on it, too.
I can’t believe I’m the only one who thinks Midnight Cowboy is a top contender. One of the greatest American movies, set almost entirely on the streets of New York. The seedy side of New York, granted, but the city and its culture is so prominent that it’s almost the third major character of the story.
Honestly, I try really hard to piss off cabdrivers as I cross the street, simply to have the chance in real life to slam a fist on their hood and yell “Hey! I’m walking here!”

It’s a crappy, sappy movie, but You’ve Got Mail really showcases all that is pleasant about the Upper West Side (I used to live there).
And they aren’t movies, but the first couple of seasons of HBO’s Sex and the City on DVD might fit your bill as well.
