Regarding Team Rocket in the Anime version of Pokémon - are James and Jessie brother and sister, or boyfriend/girlfriend?
Neither. They’re work partners and friends with some romantic tension, unless that got resolved in recent seasons. I haven’t watched in a long time, but we saw James’s family back in the…second or third season, maybe? He was a rich boy who ran away from home when he was supposed to marry a horrible woman named Jessiebell - who bore a startling resemblance to Jessie. I don’t know if we’ve ever met Jessie’s family.
In at least one of the manga, they get together in the end, and I’ve always been a bit of a Rocket-shipper, so I figure that’s the end-game, but, at least for the first few seasons, it wasn’t canon for the anime.

There’s a hint in Pokemon 2000 that James is gay (hinted in a way that was way over the kid’s heads).
