Hate to tell you this, but Naked Gun is already being re-somethinged, with Ed Helms in the Drebin role.
I’d cast Kirsten Dunst, but only as a matter of pure fanservice (the fan being me), of course she’s wrong for the part.

I think Colbert has other irons in the fire. Steve Carrell left The Office for movies. I can’t imagine him going back to series television. Except as a guest.
Ideally, you want someone well known for heavy dramatic roles who wants to exercise his comic chops. Unfortunately they’re kind of rare.
How about Mitch Pileggi? His portrayal of Skinner on “The X-Files” showed a person who can do deadpan with a bit of tongue-in-cheek when necessary. Or how about Lance Henriksen? He’s done episodic TV before in “Millenium”.
