Thanos is a supervillain whose ultimate goal is to kill half of all life in the Universe, because he’s insane and thinks that would represent some sort of “perfect balance”. He had been doing that the old-fashioned way, by raising an army and going to one planet at a time to kill a bunch of people, but that was taking too long. So he set out to collect six super-powerful Macguffin items called the Infinity Stones, and set them all in a special gauntlet so he could use all of their powers at once. The Avengers tried to stop him, but he succeeded, and accomplished his goal with a single snap of his fingers, causing a whole bunch of people (including a bunch of superheroes) to crumble into dust. Now, in Avengers: Endgame, the heroes are trying and presumably succeeding at (somehow) undoing that.
